A STUDY ON THE PREVENTIVE EFFECT OF KAM DOO DECOCTION ON THE ACUTE DDVP TOXICITY IN MOUSE


Abstract
To study on the preventive effect of Kam. Doo Decoction on the acute DDVP toxicity in mouse is to examine the effect of DDVP toxicity and preventive effect of Kam Doo Decoction, referenced in¢¥ the poison antidote of Kam Doo Decoction of oriental medicine book, in this experiment 500 mice of ICR strain were used.
The results obtained were as follows;
1. After injecting DDVP (33.3, 40.0mg/kg) by length of exposure time, ChE activities decrease 43.0~47.0¡Æo for all experimental-groups at one hour after exposure, and were gradually recovered in course of time. Also mild symptoms such as excessive sweating, vomiting and salivation, occur as DDVP poisoning symptoms.
2. The administration methods inducing Kam Doo Decoction poisoning are used P.O., S.C., I.P. and LV. There were no signs of death or poisoning up to 5000mg/kg for O.P., S.C., I.P. and up to 2000mg/kg for I.V. When Kam Doo Decoction was administrated in accordance with each method, we can conjecture the fact from this study that Kam Doo Decoction LD 50 is above 2000,5000mg/kg.
3. The ChE activity in control group was 291.76¡¾4.55nmol/ml/min, 139.0¢¥0¡¾19.59nmol ml/min in Group 1, 234.60-¡¾48.9lnmol/ml/min in Group ff and 281.72¡¾62.0lnmol/ml/min in Group 1. Compare to the control group with experimental group I (DDVP 40.0mg/kg), there was a very remarkable difference (p<0.01), and on the contrary, the comparison with experimental group ff (Kam Doo Decoction 500mg/kg), and with experimental group III (Kam Doo Decoction 1.000mg/kg) there was no remarkable difference. Kam Doo Decoction had a very remarkable preventive effect on DDVP Toxicity. So, there was able to know that ChE activity dramatically increased according to the increase in the dosage.
4. When Kam Doo Decoction for DDVP was administered by P.O. and IN. at the same time, LD 50 and confidence intervals of DDVP were as follows; the control group was 75. 9mg/kg, 65.7~83.7mg/kg, group I and II (Kam Doo Decoction 500, 1000mg/kg) by P.O. 84.1mg/kg, 68.7~102.6mg/kg, and 96.6mg/kg, 82.0~l13.8mg/kg, respectively, in the comparison with the control group, the protective index was respectively 1. 1 and 1.3. Also, in IN. administered group I and 1V (Kam Doo Decoction 750, 1000mg/kg) 106. lmg/kg, 97. 6-120.6mg/kg and 133.7mg/kg, 121.4-142.8mg/kg and a protective index of 1.4 and 1.8 we produced respectively. A protective index of Kam Doo Decoction in mouse with acute DDVP toxicity appeared and this index was higher for I.V. compared to P.O. From the above results, Kam Doo Decoction has reactivation effect of decreasing CHE activity induced by exposure to DDVP, an organophosphate pesticide.
Furthermore, Kam Doo Decoction showed a preventive effect on acute DDVP toxicity.
